Studies with cyclic adenosine monophosphate receptor and stimulation of in vitro transcription of the Gal operon.

چکیده
To elucidate further how cyclic adenosine 3’ : S’-monophosphate (cyclic AMP) and the cyclic AMP receptor are able to stimulate gal mRNA synthesis we have utilized an antiserum specific for cyclic AMP receptor and also cyclic GMP as inhibitors of cyclic AMP and cyclic AMP receptor action. When cyclic AMP, cyclic AMP receptor, DNA, and RNA polymerase are incubated together a rifampicin-resistant preinitiation complex is formed and, upon the addition of missing ribonucleoside triphosphates or Mg2+, gal mRNA synthesis occurs. If the preinitiation complex is formed in the absence of nucleoside triphosphates the antiserum is able to dissociate the complex. If the complex is formed in the presence of nucleoside triphosphates (no Mgz+), it is converted to a more stable form which is not reversed by the addition of antiserum. Cyclic GMP can prevent the formation of the preinitiation complex. However, we find that once the preinltiation complex has been formed cyclic GMP does not inhibit cyclic AMP receptor-cyclic AMP-dependent transcription, suggesting that RNA polymerase stabilizes cyclic AMP receptor-cyclic AMP binding to the gal promoter region.

Interaction of cyclic AMP receptor protein with the ilvB biosynthetic operon in E. coli.
DNase and restriction site protection studies show that cAMP and its receptor protein (CRP) bind to the promoter of the ilvB operon at approximately position -44 to -82. This region contains sequences that are homologous to those found in other CRP-dependent promoters. In vitro transcription from the ilvB promoter was markedly increased by the addition of cAMP and CRP. This stimulation was not ...
متن کاملRole of cyclic adenosine 3',5'-monophosphate and the cyclic adenosine 3',5'-monophosphate receptor protein in the initiation of lac transcription.
Optimal transcription of the lac operon in a defined purified in vitro system is dependent upon the concentrations of cyclic adenosine 3’,5’-monophosphate (cyclic AMP), the cyclic AMP-binding protein (CRP) and RNA polymerase. The concentration of CRP required for maximal Zac transcription varies proportionately with the concentration of lac DNA used as template. Maximal Zac transcription is obt...
متن کاملTranscription initiation at the Escherichia coli galactose operon promoters in the absence of the normal -35 region sequences.
The gal operon regulatory region contains two overlapping promoters, P1 and P2, regulated by cyclic AMP and the cyclic AMP receptor protein (cAMP X CRP). Starting with a mutation that eliminated P1, the promoter that is usually dependent on cAMP X CRP, we constructed a series of deletions that substituted increasing amounts of DNA sequence from upstream of P2, the promoter that usually function...
متن کاملcAMP-Epac Pathway Stimulation Modulate Connexin-43 and MicroRNA-21 Expression in Glioma Cells
Introduction: Malignant astrocytic gliomas are the most common and lethal brain malignancies due to their refractory to the current therapies. Nowadays, molecular targeted therapy has attracted great attention in treatment of glioma. Connexin 43 (Cx43) and micro ribonucleic acid- 21(miR-21) are among molecules that are involved in glioma development and progression. These molecules showed...
متن کاملInteractions of the Catabolite Activator Protein (CAP) at the Galactose and Lactose Promoters of Escherichia coli Probed by Hydroxyl Radical Footprinting
The catabolite activator protein (CAP) binding sites of the Escherichia coli galactose and lactose operons were probed by hydroxyl radical footprinting. This method reveals each base that is protected by the bound protein. The patterns of protection seen for the primary CAP sites at gal and lac were virtually identical. In the presence of RNA polymerase the footprint of the second CAP molecule ...
